A practical reference on freeze-thaw cycle: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Peptide bonds are vulnerable to protease attack, and Selank is no exception. Measured half-life in serum is short, on the order of minutes in several reports, which explains why intranasal administration is the common route described in the literature. Absorption across the nasal mucosa partially bypasses first-pass hepatic metabolism. Quantitative data on human bioavailability remain limited and are difficult to compare across studies.
Lyophilised material kept dry at minus 20 degrees Celsius or colder is the most stable form, and suppliers commonly state a shelf life of two years or more under those conditions. Once dissolved, degradation accelerates through hydrolysis and deamidation, particularly at alkaline pH or elevated temperature. Working solutions are usually divided into single-use aliquots to avoid repeated freeze-thaw cycles. The choice of reconstitution solvent affects both stability and the ionic strength of the final preparation.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres is the standard purity method. Mass spectrometry, typically electrospray ionisation, confirms identity through the expected mass-to-charge pattern. Amino acid analysis can verify composition independently. Chiral purity requires separate techniques such as derivatisation followed by chromatographic separation, and such data are rarely reported for research-grade material.
Clinical evidence comes mainly from small studies conducted in Russia, several of which were open-label or lacked robust blinding. Reported outcomes include lower anxiety scores, changes in attention measures, and effects on asthenic states following illness. Sample sizes are typically in the tens of participants, and independent replication outside the region is scarce. Reviews published in English generally note the limited methodological quality of the underlying trials. Whether the compound produces clinically meaningful effects under rigorous conditions remains unresolved.
Proposed mechanisms center on modulation of the GABA system, but no single molecular target has been confirmed. Rodent studies report changes in GABA-A receptor expression and in the turnover of serotonin, dopamine, and norepinephrine in several brain regions. Increases in brain-derived neurotrophic factor and its receptor have also been described after repeated administration. These findings come largely from animal models, and the degree to which they describe human neurochemistry remains an open question. The mechanism is best characterized as multi-system and not fully resolved.
Pharmacokinetic data are limited. Like most short peptides, Selank is vulnerable to plasma and tissue peptidases, and its measured half-life in circulation is short, on a minutes scale. The Pro-Gly-Pro tail slows this degradation but does not eliminate it. Intranasal administration is the route described in most reports, with absorption through the nasal mucosa and a hypothesized path into the central nervous system that avoids the blood-brain barrier. Direct measurements of human brain exposure are unavailable, so distribution claims rest on inference from animal work.

The primary structure of Selank is Thr-Lys-Pro-Arg-Pro-Gly-Pro, corresponding to the molecular formula C33H57N11O9 and a monoisotopic mass of roughly 751.9 daltons. The N-terminal threonine and the arginine residue in the fourth position are shared with tuftsin, which carries the sequence Thr-Lys-Pro-Arg. The three additional residues at the C-terminus, Pro-Gly-Pro, extend the chain and are associated with greater resistance to enzymatic degradation. This extension also separates Selank from the shorter parent peptide.
Naming conventions place Selank in the same research family as Semax, another Russian-developed peptide investigated for cognitive effects. The two compounds share a lineage but differ in sequence and in the biological systems proposed as their targets. Semax descends from ACTH fragments, whereas Selank descends from tuftsin. Publications sometimes identify Selank by its full peptide sequence or by laboratory codes rather than one uniform trade name. Because replication outside Russia is limited, reports on its properties are best read alongside the study design and the purity of the material tested.
Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Photoanomerization with UV light allows for inversion about the 1' anomeric centre to give the correct beta stereochemistry; one problem with this chemistry is the selective phosphorylation of alpha-cytidine at the 2' position. However, in 2009, they showed that the same simple building blocks allow access, via phosphate controlled nucleobase elaboration, to 2',3'-cyclic pyrimidine nucleotides directly, which are known to be able to polymerise into RNA. Organic chemist Donna Blackmond described this finding as "strong evidence" in favour of the RNA world. However, John Sutherland said that while his team's work suggests that nucleic acids played an early and central role in the origin of life, it did not necessarily support the RNA world hypothesis in the strict sense, which he described as a "restrictive, hypothetical arrangement". The Sutherland group's 2009 paper also highlighted the possibility for the photo-sanitization of the pyrimidine-2',3'-cyclic phosphates. A potential weakness of these routes is the generation of enantioenriched glyceraldehyde, or its 3-phosphate derivative (glyceraldehyde prefers to exist as its keto tautomer dihydroxyacetone). On August 8, 2011, a report, based on NASA studies with meteorites found on Earth, was published suggesting building blocks of RNA (adenine, guanine, and related organic molecules) may have been formed in outer space.
Spondyloepiphyseal dysplasia congenita is one of a spectrum of skeletal disorders caused by mutations in the COL2A1 gene. The protein made by this gene forms type II collagen, a molecule found mostly in cartilage and in the clear gel that fills the eyeball (the vitreous). Type II collagen is essential for the normal development of bones and other connective tissues. Mutations in the COL2A1 gene interfere with the assembly of type II collagen molecules, which prevents bones from developing properly and causes the signs and symptoms of this condition. Spondyloepiphyseal dysplasia congenita is inherited in an autosomal dominant pattern, which means one copy of the altered gene is sufficient to cause the disorder.

=== Organoindium compounds === Organoindium compounds feature In–C bonds. Most are In(III) derivatives, but cyclopentadienylindium(I) is an exception. It was the first known organoindium(I) compound, and is polymeric, consisting of zigzag chains of alternating indium atoms and cyclopentadienyl complexes. Perhaps the best-known organoindium compound is trimethylindium, In(CH3)3, used to prepare certain semiconducting materials.
== Ethics == AI has potential benefits and potential risks. AI may be able to advance science and find solutions for serious problems: Demis Hassabis of DeepMind hopes to "solve intelligence, and then use that to solve everything else". However, as the use of AI has become widespread, several unintended consequences and risks have been identified. In-production systems can sometimes not factor ethics and bias into their AI training processes, especially when the AI algorithms are inherently unexplainable in deep learning.

=== Dentistry === Calcium sulfate has a long history of use in dentistry. It has been used in bone regeneration as a graft material and graft binder (or extender) and as a barrier in guided bone tissue regeneration. It is a biocompatible material and is completely resorbed following implantation. It does not evoke a significant host response and creates a calcium-rich milieu in the area of implantation.
The Shine–Dalgarno (SD) sequence is, sometimes partially, part of a ribosomal binding site in bacterial and archaeal messenger RNA. It is generally located around 8 bases upstream of the start codon AUG. The RNA sequence helps recruit the ribosome to the messenger RNA (mRNA) to initiate protein synthesis by aligning the ribosome with the start codon. Once recruited, tRNA may add amino acids in sequence as dictated by the codons, moving downstream from the translational start site. The Shine–Dalgarno sequence is common in bacteria, but rarer in archaea. It is also present in some chloroplast and mitochondrial transcripts. The six-base consensus sequence is AGGAGG; in Escherichia coli, for example, the sequence is AGGAGGU, while the shorter GAGG dominates in E. coli virus T4 early genes. The Shine–Dalgarno sequence was proposed by Australian scientists John Shine and Lynn Dalgarno in 1973.
American black bears were once not considered true or "deep" hibernators, but because of discoveries about the metabolic changes that allow black bears to remain dormant for months without eating, drinking, urinating or defecating, most biologists have redefined mammalian hibernation as "specialized, seasonal reduction in metabolism concurrent with scarce food and cold weather". American black bears are now considered highly efficient hibernators. The physiology of American black bears in the wild is closely related to that of bears in captivity. Understanding the physiology of bears in the wild is vital to the bear's success in captivity. The bears enter their dens in October and November, although in the southernmost areas of their range (i.e. Florida, Mexico, the southeastern United States), only pregnant females and mothers with yearling cubs will enter hibernation. Prior to that time, they can put on up to 14 kg (30 lb) of body fat to get them through the several months during which they fast. Hibernation typically lasts 3–8 months, depending on regional climate. Hibernating bears spend their time in hollowed-out dens in tree cavities, under logs or rocks, in banks, caves, or culverts, and in shallow depressions. Although naturally-made dens are occasionally used, most dens are dug out by the bear. During their time in hibernation, an American black bear's heart rate drops from 40 to 50 beats per minute to 8 beats per minute, and the metabolic rate can drop to a quarter of the bear's (non-hibernating) basal metabolic rate.

Dry powder is best kept sealed, protected from light, and held at minus 20 degrees Celsius or below. Desiccant packaging helps limit moisture uptake because the material is hygroscopic. A sealed vial should be allowed to equilibrate to room temperature before opening to reduce condensation.
Hydrolysis of peptide bonds and deamidation of arginine and proline residues are the main degradation routes. Alkaline pH and higher temperatures accelerate both processes. Buffered, near-neutral solutions stored cold generally degrade more slowly than unbuffered ones.
Reverse-phase chromatography gives the clearest single view of related impurities and truncated sequences. Mass spectrometry then confirms that the main peak carries the expected mass. Neither test detects enantiomeric impurities, which require a separate chiral assay.
Reports describe effects on GABA-A receptor expression, monoamine turnover, and neurotrophic factor levels. These are proposed mechanisms drawn mainly from animal models. No single molecular target has been established.
