Proposed mechanisms center on modulation of the GABA system, but no single molecular target has been confirmed. Rodent studies report changes in GABA-A receptor expression and in the turnover of serotonin, dopamine, and norepinephrine in several brain regions. Increases in brain-derived neurotrophic factor and its receptor have also been described after repeated administration. These findings come largely from animal models, and the degree to which they describe human neurochemistry remains an open question. The mechanism is best characterized as multi-system and not fully resolved.
Pharmacokinetic data are limited. Like most short peptides, Selank is vulnerable to plasma and tissue peptidases, and its measured half-life in circulation is short, on a minutes scale. The Pro-Gly-Pro tail slows this degradation but does not eliminate it. Intranasal administration is the route described in most reports, with absorption through the nasal mucosa and a hypothesized path into the central nervous system that avoids the blood-brain barrier. Direct measurements of human brain exposure are unavailable, so distribution claims rest on inference from animal work.
Clinical evidence comes mainly from small studies conducted in Russia, several of which were open-label or lacked robust blinding. Reported outcomes include lower anxiety scores, changes in attention measures, and effects on asthenic states following illness. Sample sizes are typically in the tens of participants, and independent replication outside the region is scarce. Reviews published in English generally note the limited methodological quality of the underlying trials. Whether the compound produces clinically meaningful effects under rigorous conditions remains unresolved.
Development took place at the Institute of Molecular Genetics of the Russian Academy of Sciences, where a series of short peptides were designed in the 1980s and 1990s. Selank was selected from variants of tuftsin that showed resistance to plasma peptidases. Russian regulatory approval covers it as an anxiolytic agent given intranasally. Outside that market the compound is normally handled as a research chemical rather than a medicine, and no widely recognised international pharmacopoeial monograph exists. The name Selank is a coined trade designation rather than a systematic chemical name.
Enzymatic stability motivates the extra three residues at the carboxyl end. Native tuftsin is cleaved quickly by circulating aminopeptidases and carboxypeptidases, which limits its duration of action and its usefulness as a tool compound. Extending the chain with proline-rich segments is a common design tactic because proline constrains the backbone and slows proteolysis. The same Pro-Gly-Pro motif appears in other Russian-developed peptides of the era. Whether the full seven-residue chain is required for activity, or whether it acts mainly as a prodrug releasing tuftsin, remains unresolved.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. Its four N-terminal residues reproduce tuftsin, a tetrapeptide fragment of the immunoglobulin heavy chain, while the C-terminal Pro-Gly-Pro extension is a synthetic addition. The peptide has a molecular mass near 752 daltons and carries a net positive charge at physiological pH because of the arginine and lysine side chains. Published indexes list it under the name Selank and the sequence abbreviation TKPRPGP. Solid-phase peptide synthesis is the standard production route for research quantities.

Selank is not a naturally occurring peptide and has no known endogenous counterpart in human physiology. Russian-language sources frequently call it TP-7, while English-language sources use the name Selank almost exclusively. Database indexing is uneven, partly because early reports appeared in regional journals that are not widely cataloged. Some summaries describe the material as a tuftsin analog and others as a synthetic heptapeptide; the labels overlap rather than conflict. Citing the primary sequence resolves ambiguity more reliably than the research or trade name alone.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, written TKPRPGP in one-letter notation. Its structure consists of the immunomodulatory tetrapeptide tuftsin, Thr-Lys-Pro-Arg, extended at the carboxyl terminus by a Pro-Gly-Pro segment. The molecular formula is commonly given as C33H57N11O9, corresponding to a monoisotopic mass near 751.4 Da and an average molecular mass near 751.9 Da. All seven residues are proteinogenic amino acids, and the molecule carries no modified side chains or non-natural linkages.
The compound was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences during the 1980s and 1990s. The stated design goal was to retain the immunomodulatory and central nervous system activity attributed to tuftsin while improving resistance to enzymatic breakdown. Adding a proline-rich tail to the short parent peptide was a deliberate strategy, because proline residues restrict the conformations available to many peptidases. The same laboratory produced Semax, an ACTH fragment analog, and both compounds were developed in parallel as short, enzymatically stabilized peptides intended for intranasal use.
Dry powder is normally held at -20 degrees Celsius or lower, in a sealed container with desiccant and protection from light. Reconstituted solutions are usually kept at 2 to 8 degrees Celsius for short periods and frozen for longer ones. Proline residues at several positions are generally associated with some resistance to peptidase attack, but chemical stability still declines at neutral to alkaline pH and at elevated temperature. Exact shelf-life figures are product-specific and are not standardised across suppliers.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, with mass spectrometry used to confirm molecular mass and sequence information. Amino acid analysis and peptide mapping may supplement these methods. Certified reference standards are scarce, and many commercial lots are sold as research chemicals without a pharmacopoeial monograph. Regulatory treatment differs by country: Selank is a registered prescription medicine in Russia, while in the European Union and the United States it is not an approved drug and may fall under research-chemical or unapproved-product frameworks.
==== Edema and connective tissue changes ==== As the blood-nerve-barrier breaks down, proteins and cells will be able to enter the perineural and endoneurial space. The increased permeability allowing substances to enter combined with the lack of a lymphatic system to drain fluids causes an increase in pressure and may alter the ionic environment. The increased pressure in the endoneurium can cause a mini "compartment syndrome" leading to post-traumatic ischemia of the nerve cells. When endoneurial edema is triggered, the swelling will last many hours the point that the entrapment is relieved. For example, following 2-8 hours of compression, endoneurial fluid pressure will rapidly rise and can stay elevated for 24 hours. Lymphocytes, fibroblasts, and macrophages will also be able to cross the newly permeable blood nerve barrier and react to the antigens contained in the perineural space triggering an inflammatory reaction. As part of this inflammatory reaction, there will be excess deposits of fibrin (i.e. scarring). On histological analysis, epineural fibrosis and perineural thickening can be seen. This scarring is an irreversible change associated with nerve entrapment. If the scarring damages the microcirculatory environment, then the impaired blood supply will also be permanent. In cases where permanent impairment exists even after a nerve decompression, it's thought that the pathophysiological basis is due to extensive scarring along and within the nerve, as demyelination and axonmetesis are generally capable of healing but scarring cannot be reversed.
It was first advertised on television with the jingle "Brylcreem — A Little Dab'll Do Ya! Brylcreem — You'll look so debonair. Brylcreem — The gals'll all pursue ya; they'll love to run their fingers through your hair!". Another version was "Brylcreem—a little dab will do ya! Use more only if you dare; but watch out! The gals will all pursue ya! They'll love to run their fingers through your hair!" The jingle was created by Hanley M. Norins of the Young & Rubicam advertising agency. The television advertisement for Brylcreem included a cartoon animation of a man with (initially) shaggy hair, who happily has a little dab applied, and, miraculously, the hair combs and smooths itself. When the dry look became popular, partly inspired by the unoiled moptops of the Beatles, the last line was changed from "They'll love to run their fingers through your hair", to "They'll love the natural look it gives your hair". Subsequent television advertisements used the mottoes "Grooms without gumming" and later, in the 1970s, in the United Kingdom and Canada, "A little dab of Brylcreem on your hair gives you the Brylcreem bounce".
Also, during the 1981 tour, veteran session drummer Andy Newmark stepped in briefly for Knudsen, who was then in rehab. And by the end of 1981, even Simmons had left the band. Now with no remaining original members and a "leader" in McDonald who was ready for a solo career, the group elected instead to disband after a rehearsal without Simmons, according to an interview with McDonald for Listen to the Music, the Doobie Brothers' official video history/documentary released in 1989. He went on to say that by that point they could not have gotten further away from the Doobies sound if they had tried. Simmons, already at work on his first solo album, Arcade, rejoined for a 1982 farewell tour on the condition that this truly would be the end of the Doobie Brothers. At their last concert at the Greek Theatre in Berkeley, California, on September 11, 1982, they were joined onstage by founding member Tom Johnston for what was presumed to be the final rendition of his staple, "China Grove". Former members Porter, Hossack and Hartman subsequently took the stage for an extended version of "Listen to the Music". Knudsen sang lead vocals while Johnston, Simmons and McFee traded licks on guitar. The live album Farewell Tour was released in 1983 and the Greek Theatre concert was released in 2011 as Live at the Greek Theatre 1982.

=== Clinical research === The therapeutic effects of bromantane in asthenia are said to onset within 1–3 days. It has been proposed that the combination of stimulant and anxiolytic activity may give bromantane special efficacy in the treatment of asthenia. In a large-scale, multi-center clinical trial of 728 patients diagnosed with asthenia in Russia, bromantane was given for 28 days at a daily dose of 50 mg or 100 mg. The study concluded with an impression score of 76.0% on the CGI-S and 90.8% on the CGI-I for bromantane, indicating that it is broadly applicable and highly effective. The therapeutic benefit against asthenia was observed to still be present one month after discontinuation of the drug. 3% of patients experienced side effects; though none were considered serious; and 0.8% of patients discontinued treatment due to side effects. Bromantane was also noted to normalize the sleep–wake cycle.
When we were getting very close to releasing Half-Life (less than a week or so), we found there were already some projects that we needed to start working on, but we couldn't risk checking in code to the shipping version of the game. At that point we forked off the code in VSS to be both /$Goldsrc and /$Src. Over the next few years, we used these terms internally as "Goldsource" and "Source". At least initially, the Goldsrc branch of code referred to the codebase that was currently released, and Src referred to the next set of more risky technology that we were working on. When it came down to show Half-Life 2 for the first time at E3, it was part of our internal communication to refer to the "Source" engine vs. the "Goldsource" engine, and the name stuck. Source was developed part-by-part from this fork onwards, slowly replacing GoldSrc in Valve's internal projects and, in part, explaining the reasons behind its unusually modular nature. Valve's development of Source since has been a mixture of licensed middleware and in-house-developed code. Older versions of Source use Bink Video for video playback, however more recent releases of the Source engine use WebM videos for menu backgrounds, Full Motion Videos, and splash screens. .
=== Mass spectrometry === MALDI-TOF (Matrix-assisted laser desorption/ionization - time of flight) is a specific type of mass spectrometry that is able to identify microorganisms. A pure culture is isolated and spread directly on a stainless steel or disposable target. The cells are lysed and overlaid with a matrix, which forms protein complexes with the bacterial proteins. The MALDI fires a laser and ionizes the protein complexes, which break off and travel up the vacuum where they are detected based on mass and charge. The resulting protein spectra is compared to a known database of previously catalogued organisms, resulting in rapid diagnosis of microorganisms. Recent studies have suggested that these tests can become specific enough to diagnose down to the sub-species level by observing novel biomarkers. The MALDI-TOF identification method requires pure cultures that are less than 72 hours old. This places the organism in log phase with an abundance of ribosomal proteins, which are the most common proteins detected in the spectra. Identifications with this technology can also be impacted if the culture is exposed to cold temperatures, as this would change the typical protein distribution.
The widespread perception about the economic management of Rodríguez Zapatero's government during the "legislature of crisis" was that it had failed, despite having managed to avoid the European bailout. That perception was key to the People's Party's landslide victory in the general election of November 2011. The deep economic crisis translated into a political crisis from the moment that the lack of confidence in the government's ability to deal with it was transferred to the entire "political class" and the system as a whole. To this was added the proliferation of corruption scandals involving the two main parties — Gürtel affair, Palma Arena case, Andalucian ERE affair — and even the Royal House when the King's son-in-law Iñaki Urdangarín was indicted in 2011, an affair that had a huge media repercussion and deteriorated the image of the monarchy. The rating of the government, its president and the PSOE were falling in the polls and in the Galician regional election of 2009 and in the European Parliament elections of the same year, the Socialists were defeated. The fall was accentuated after the economic policy turnaround of May 2010. The first confirmation of the Socialist slump came in the Catalan elections of November 2010 in which the Socialists' Party of Catalonia lost nine deputies, and the "three party" was ousted from power by CiU – the "convergent" Artur Mas replaced the socialist José Montilla at the head of the Generalitat.

Reports describe effects on GABA-A receptor expression, monoamine turnover, and neurotrophic factor levels. These are proposed mechanisms drawn mainly from animal models. No single molecular target has been established.
Intranasal delivery is the route described in most published work. It is used because the peptide is degraded quickly once it reaches circulation. Direct evidence of brain penetration in humans is lacking.
Most trials are small, regionally concentrated, and often lack rigorous blinding or placebo control. Independent replication is limited. Reviews in English generally rate the evidence as preliminary.
Selank is a seven-amino-acid peptide built from the tuftsin sequence plus a Pro-Gly-Pro tail. It is produced by chemical synthesis rather than extracted from a natural source. The free peptide is usually supplied as a lyophilised powder or in an aqueous formulation.
